The Arsenal goalkeeper managed a light training session on Wednesday, but England boss Sven Goran Eriksson has today decided that he will not be fit enough to play at Old Trafford.
Seaman felt another twinge in his right shoulder as he trained, dashing initial hopes that he would be able to take his place in the side. Leeds keeper Nigel Martyn is poised to step into the breach.
In addition, Leicester keeper Ian Walker has been called into the squad as cover for Martyn and Arsenal's Richard Wright.
Eriksson said: 'It is obviously disappointing for David that he will miss Saturday's final qualifying match against Greece.
'However, we have other first-class goalkeepers in the squad in Nigel Martyn, Richard Wright and now Ian Walker.
'I'm confident that whoever we select to play against Greece will do an excellent job for England.'
England are already without Michael Owen, Sol Campbell, Wes Brown
and Alan Smith through injury, while Nicky Butt's fitness is to be assessed on Friday following further training.
Wenger had already warned that Seaman's fitness could be a doubt, saying: 'He has had some inflammation in the shoulder for a while and he told me it has slowly got worse. We need to be careful with it - we need some medical advice about him.'
The shoulder problem had kept Seaman out of Arsenal's 2-0 Premiership win at Derby last Saturday.
